The Mechanisms Behind Lipid Nanoparticles
Lipid Nanoparticle technology has proven to be essential for the delivery of mRNA vaccines and is emerging as the prominent vehicle to deliver a variety of therapeutics. With its focus on immunologic indications (i.e. oncology and infectious disease vaccines), Providence Therapeutic has taken a different view on developing ionizable lipids and LNP formulations.
Providence Therapeutics' philosophy on Lipid Nanoparticles is that there is no LNP that can do it all. That is why we are developing multiple LNP families. Some members of these families are optimized for stimulation of the cytotoxic T cell required for tumour cell destruction for maximizing cancer vaccine efficacy or attacking latent viruses. While other families are optimized for the generation of neutralizing antibodies needed to prevent infectious diseases such as influenza, COVID, and traveller’s diseases.
Integrating Lipid Nanoparticles in Healthcare
We have designed 250 ionizable lipids using rational design principles centred around enhancing the specific arms of the immune system. These ionizable lipids are used to make different LNPs with formulations chosen for stability, high-level mRNA delivery, minimal toxicity, and ease of manufacture. The screening of all 250 lipids is ongoing, but we have already identified multiple lipids and LNP formulations that have been chosen as our next-generation mRNA-LNPs for oncology and infectious disease use.

Providence is focused on oncology and infectious diseases, but during our ongoing screening, it is also searching for next-generation lipids and LNP that will be applicable for gene therapy applications (i.e. high level of gene delivery or specific cell delivery, with only minimal immune activation) or that demonstrate enhanced mixed immunologic responses that would apply & to bacterial and parasitic infections.
